 you can enter after waters of life, but u cant join untill SPOILER ALERT!!!!! you get kidnapped by the Enclave, then go to the pentagon, and you are given a choice to join Lyon's Pride, but your almost done with the game by then

 be nice and go underground and kill his heart, make him grow more with the stuff mother maple gives you, make him stop growing with the stuff Father Poplar gives you, or be evil and torch him with a flame or shishkebab

 2 ways- 1 go to your map and look down the river for the pentagon, or 2 SPOILER ALERT!!! BTW, you can only enter by going through the story, other wise, they wont let u inside go through the main storyline, aft..

 look for the little panel on the left side of the door, you'll go inside and see officer gomez, and he'll tell u about the vaults problems
